15-P002 Limbs gone batty: A second wave of Sonic hedgehog expression during the development of the bat limb

چکیده
The first lineage allocation during mouse development results in the trophectoderm and inner cell mass at the blastocyst stage. The caudal-related transcription factor, CDX2, is upregulated and required for the repression of inner cell mass genes Oct 4 and Nanog in the trophectoderm to specify the two lineages. Mouse embryos are precocious implanters requiring early establishment of trophectoderm. It is unclear whether this is conserved between mammals, particularly in embryos that implant late during embryonic development, such as marsupial embryos. Using nested PCR, we have cloned a partial CDX2 sequence from the stripe-faced dunnart (Sminthopsis macroura) based on known mammalian CDX2 sequences. Dunnart CDX2 shares 97.8% and 86.7% sequence homology to the gray short-tailed opossum and mouse CDX2 sequence, respectively, suggesting conserved homology among mammalian species. Using gene expression analysis, dunnart CDX2 is expressed in the gut and the ovary, consistent with studies in mice. However, unlike in mice when CDX2 can be detected as early as the 8-cell stage, dunnart CDX2 is not expressed during early embryonic development until the late unilaminar blastocyst stage, Day 5.5 of a 10.7 day gestation period. This difference in expression may be related to later trophectoderm differentiation, hypoblast allocation and implantation in the dunnart. This is the first study of CDX2 in a marsupial species and suggests that CDX2 expression is involved in lineage determination in the dunnart.

A second wave of Sonic hedgehog expression during the development of the bat limb.
Sonic hedgehog (Shh) plays an integral role in both the anterior-posterior (A-P) patterning and expansion of developing vertebrate limbs through a feedback loop involving Fgfs, Bmps, and Gremlin. In bat limbs A-P patterning and the size of the digital field are unique. The posterior digits of the forelimb are elongated and joined by tissue, whereas the thumb is short. The hindlimb digits often ...
متن کاملThe role of the desert hedgehog signaling pathway during degeneration and regeneration of peripheral nerves
The desert hedgehog (Dhh) signaling pathway is involved in the development of peripheral nerves (PNs). Dhh-null mice show abnormal neuronal development and perineurial barrier function. As it was previously shown that dhh is mainly expressed in developmental nerves and Sonic hedgehog protein (dhh homologous) has therapeutic effects in neuronal survival, we attempted to investigate the possible ...

Gastric cancer (GC) is considered as one of the most serious cancers, and in Iran, due to environmental factors in different regions, it has a high frequency. Apart from environmental factors, genetic and epigenetic ones also play a key role in the development of carcinogenesis of GC. In this regard, the study of functioning of the molecular mechanisms involved in the carcinogenesis and also tu...
متن کاملConservation in hedgehog signaling: induction of a chicken patched homolog by Sonic hedgehog in the developing limb.
Hedgehog genes have been implicated in inductive signaling during development in a variety of organisms. A key element of the hedgehog signaling system is encoded by the gene patched. In Drosophila hedgehog regulates gene expression by antagonizing the action of patched. In addition, patched is itself a transcriptional target of hedgehog signaling. We have isolated a chicken patched homolog and...
